近期,中国医学科学院皮肤病研究所姚煦教授团队、复旦大学附属华山医院李巍教授团队及香港城市大学董立新教授团队合作在 Cell Host & Microbe(IF=20.6)上发表题为“Multi-omics Signatures Reveal Genomic and Functional Heterogeneity of Cutibacterium acnes in Normal and Diseased Skin.”(多组学特征揭示正常和患病皮肤中痤疮丙酸杆菌的基因组和功能异质性)的研究,在该项目中,研究团队从1234个分离基因组中证明了C. acnes的个体和生态位异质性。皮肤疾病(湿疹和痤疮)和身体部位决定了C. acnes的基因组差异,这种差异源于水平基因转移和选择压力。与表皮葡萄球菌(Staphylococcus epidermidis)相比,C. acnes具有独特的代谢功能,抗生素耐药基因和毒力因子较少,基因组更稳定。综合基因组、转录组和代谢组分析在菌株水平揭示了C. acnes的功能特征。与转录组特征一致,C. acnes在富含皮脂的环境中对角质形成细胞产生毒性和促炎作用。L-肌肽(L-carnosine)是一种抗氧化应激代谢物,在湿疹的代谢组中上调并减轻皮肤炎症。总的来说,该研究揭示了基因和微环境对C. acnes功能的共同影响。利用从健康皮肤、湿疹(AD)和痤疮患者皮肤分离的痤疮丙酸杆菌(Cutibacterium acnes, C. acnes)菌株进行了全基因组测序、转录组测序、代谢组测序以及功能研究,研究团队首次从单菌株层面揭示了C. acnes的基因和功能异质性。通过深入地了解宿主和皮肤微生物群之间的复杂关系,有望能找到增进皮肤健康和改善皮肤病管理的靶点,也为将来探索C. acnes的临床应用奠定了基础。
