耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)感染引起的骨髓炎很难根除,甚至会危及生命。考虑到感染是持久的并且深入骨组织,骨髓炎的控制和有效治疗仍然具有挑战性。最近,南开大学的研究团队在期刊《Advanced Materials》(IF=32.086)上发表题为“Guanidinium‐Decorated Nanostructure for Precision Sonodynamic‐Catalytic Therapy of MRSA‐Infected Osteomyelitis.”的文章,提出了一种可活化的纳米结构(Au/TNT@PG ),用于MRSA感染的骨髓炎的协同声动力-催化治疗。Au/TNT@PG主链是通过将富含胍盐的聚合物(PG)结合到吸收超声波(US)的掺杂金的钛酸盐纳米管(Au/TNTs)上而获得的,该聚合物是渗透生物膜基质的成分。在深穿透超声波作用下,纳米复合材料产生1O2用于声动力治疗(SDT ),并在酸性感染微环境中催化内源性H2O2分解为有毒的·OH用于催化治疗,导致细菌细胞死亡。其强大的抗菌效果归因于其细菌捕获能力、带正电荷的胍盐的生物膜渗透性以及随后的Au/TNT的声动力-催化作用的协同效应。这种远程控制的方法增强了巨噬细胞向M2型的极化,同时抑制了M1型,导致局部炎症消退并增强了成骨细胞增殖和分化,从而抑制了骨丢失。这项研究为骨髓炎的有效声动力催化治疗提供了一种通用的纳米治疗方法。
本文中使用了欣博盛Rat IL-1β, TNF-α, IL-4 and IL-10 ELISA kits检测了骨髓组织匀浆样本中这些细胞因子的含量。
