特发性肺纤维化(IPF, Idiopathic pulmonary fibrosis)是一种慢性进行性间质性肺病,最终会导致肺功能永久恶化并死亡。白细胞介素-11 (IL-11)是一种促纤维化细胞因子,对成纤维细胞分化为分泌胶原、肌动蛋白α2、平滑肌阳性(ACTA2+)肌成纤维细胞至关重要,是特发性肺纤维化(IPF)发病机制的驱动过程。最近,上海交通大学药学院在期刊《Science Advances》(IF=14.957)上发表题为“Inhaled siRNA nanoparticles targeting IL11 inhibit lung fibrosis and improve pulmonary function post-bleomycin challenge.”的文章,开发了一种可吸入和可粘液渗透的纳米颗粒(NP)系统,该系统结合了抗IL11的siRNA(siIL11 @ PPGC NPs ),并研究了其治疗IPF的治疗潜力。NP是通过可生物降解的PLGA-PEG二嵌段共聚物和自创的阳离子类脂分子G0-C14的自组装来配制的,以实现siIL11的有效跨粘膜递送。非侵入性雾化吸入通过抑制ERK和SMAD2来阻止成纤维细胞分化和减少ECM沉积。此外,在博莱霉素诱导的肺纤维化的小鼠模型中,siIL11@PPGC NPs显著减少了纤维化的发展并改善了肺功能,而没有诱导全身毒性。这项研究为siRNA治疗剂的局部吸入给药提供了一个通用的纳米颗粒平台,并在许多呼吸系统疾病(包括IPF)的治疗中显示出有希望的临床潜力。
本文中使用了欣博盛Mouse TGF-β1 ELISA kit检测了肺泡灌洗液样本中的表达量。
